Animal reproduction studies have shown an adverse effect on the fetus and there are no adequate and well-controlled studies in humans, but potential benefits may warrant use of the drug in pregnant women despite potential risks.
There is positive evidence of risk to a breastfed infant or to breastmilk production, but the benefits of use in breastfeeding mothers may be acceptable despite the risk to the infant (e.g. if the drug is needed in a life-threatening situation or for a serious disease for which safer drugs cannot be used or are ineffective).
Typical usage: Schizophrenia, affective withdrawal and social maladjustment.
Side Effects: Lower threshold for seizures, blurring of vision, dry mouth, retention of urine, constipation, orthostatic hypotension, weight gain, impaired glucose tolerance, allergic skin rashes, cholestatic jaundice; extrapyramidal effects; delirium; agitation, anxiety, depression, euphoria.
Drug Interaction: Orthostatic hypotension with MAOIs; may increase sedation with alcohol, hypnotics, antihistamines, opiates; antacids containing aluminum salts may decrease absorption; additive antimuscarinic effects with TCAs; may reduce bromocriptine's ability to reduce serum prolactin; amphetamines may increase psychosis.
Mechanism Of Action: Penfluridol blocks the postsynaptic dopamine receptor in the mesolimbic dopaminergic system and inhibits the release of hypothalamic and hypophyseal hormones.
