Studies in breastfeeding mothers have demonstrated that there is significant and documented risk to the infant based on human experience, or it is a medication that has a high risk of causing significant damage to an infant. The risk of using the drug in breastfeeding women clearly outweighs any possible benefit from breastfeeding. The drug is contraindicated in women who are breastfeeding an infant.
Typical usage: menopause, treating vulval or vaginal atrophy, prostate cancer
Side Effects: Gynacomastia, uterine bleeding, breast cancer, endometrial carcinoma, headache, depression, weight gain, nausea and vomiting, fluid retention, breast tenderness.
Drug Interaction: Estradiol. is known to interact with other drugs like phenytoin (Na), rifampicin, sulphamethizole. Always consult your physician for the change of dose regimen or an alternative drug of choice that may strictly be required.
Mechanism Of Action: Estradiol enters target cells freely (e.g., female organs, breasts, hypothalamus, pituitary) and interacts with a target cell receptor. When the estrogen receptor has bound its ligand it can enter the nucleus of the target cell, and regulate gene transcription which leads to formation of messenger RNA. The mRNA interacts with ribosomes to produce specific proteins that express the effect of estradiol upon the target cell. Estrogens increase the hepatic synthesis of sex hormone binding globulin (SHBG), thyroid-binding globulin (TBG), and other serum proteins and suppress follicle-stimulating hormone (FSH) from the anterior pituitary.
